First, the problem of correct installation

Before using the microscope, first install the eyepiece and objective lens of the microscope. The installation of the eyepiece is relatively simple. The main problem is the installation of the objective lens. Since the objective lens is more expensive, if the student does not have the thread when installing, it is easy to fall to the ground, causing damage to the lens. Therefore, for the sake of safety, the student is emphasized when installing the objective lens. Hold the objective lens with your left index finger and middle finger, and then attach the objective lens with your right hand so that it will not fall to the ground even if it is not installed.

Second, the correct problem with light

Optics is an important step when using a microscope. Some students turn a objective lens toward the light-passing hole when they are facing the light, instead of using a low-power mirror to light the light. I like to use one hand when turning the mirror, and the mirror is often pulled down. Therefore, when the teacher is guiding the students, it is necessary to emphasize the use of low magnification mirrors for light. When the light is strong, use small apertures and plane mirrors. When the light is weak, use large apertures and concave mirrors. The mirrors should be rotated with both hands. Until a uniform bright circular field of view. Do not move the microscope casually after the light is on, so that the light does not pass through the mirror into the clear hole.

Third, the correct use of quasi-focus spiral

Using a quasi-focus spiral to adjust the focal length, finding the object can be said to be the most important step in the use of the microscope, and is the most difficult step for students. During the operation, students are prone to the following errors: First, direct focus adjustment under high power microscope; second, regardless of the rise or fall of the lens barrel, the eye always looks at the field of view in the eyepiece; third, the critical value of the object distance is not known. When the distance is adjusted to 2 to 3 cm, it is still up, and the speed of rotating the quasi-focus spiral is very fast. The first two kinds of erroneous results often cause the objective lens to interfere with the loading, damage loading or lens, and the third error is the most common phenomenon when students use the microscope. In response to the above mistakes, the teacher must emphasize to the students that the adjustment of the focal length must be adjusted at the low magnification mirror, first turn the coarse focus spiral, so that the lens barrel slowly descends, the objective lens is close to the slide, but be careful not to let the objective lens touch the glass. In this process, the eye should look at the objective lens from the side, then use the left eye to look into the eyepiece, and slowly adjust the coarse pseudo-focus spiral to make the lens tube rise slowly until the object image is seen, and explain to the students. Generally, the object distance of the microscope is about 1 cm, so if the object distance is far more than 1 cm, but the object image is still not seen, it may be that the specimen is not in the field of view or the coarse coke spiral speed is too fast. The position of the film is loaded, and then the above steps are repeated. When a blurred object image appears in the field of view, the fine focus screw adjustment is used instead. In this way, the search range can be narrowed and the speed of finding the object image can be improved.

Fourth, the problem of objective lens conversion

After using a low power mirror and switching to a high power microscope, students often prefer to use a finger to directly push the objective lens. This is considered to be labor-saving, but it is easy to deflect the optical axis of the objective lens because the material of the converter is soft and has high precision. , the thread is not evenly stressed and it is easy to loosen. Once the thread breaks, the entire converter is scrapped. The instructor should instruct the student to hold the lower rotating plate of the converter to convert the objective lens.

Fifth, the problem of correct use of the eye

When observing an object with a microscope, both eyes should be opened at the same time, and the left eye should be looked into the eyepiece. However, many students often can't do this. They like to hold their right eyes with their hands or simply close their right eyes. This is not in line with the experimental observation requirements. This bad habit can cause fatigue in the left eye and cannot be done at the same time. Draw a picture while watching. While pointing out the problem of the students, the teacher should demonstrate specifically, telling the students that the left eye should be as close as possible to the eyepiece, and the right eye should try to inspect it in the field of view. If this training is repeated, it will achieve the goal of both eyes and observation. Or you can do the following exercises: Open your eyes, use a piece of paper or palm to stand between your eyes, with your nose facing forward, so that the left and right eyes can't look at each other on the opposite side, then consciously look at the left side and then the right side. 3 to 5 times, do it every morning and evening, and learn in less than a week.

The use or operation of the microscope is a common phenomenon in biological experiments. We only need to take it seriously, consciously correct it, overcome it, and use the microscope skillfully and correctly.
